Can I close the registers of the rooms that I do not use on a regular basis?

An air conditioner is designed to cool a certain squre feet of area. If you close the registers to rooms which are not occupied this square gets reduced. As a result it has to work harder to cool a smaller area. It’s efficiency and the airflow is disturbed. It will cycle more often which is bad for its health. You will not get sufficient cooling and power bills will be higher.
